Mempro™ cell-free protein production system is the innovated system to produce transmembrane proteins, which bypasses the limitations that encountered in cell-based production system, such as host cell toxicity, low yield and solubilization and purification using detergents. Integrins are a family of cell adhesion receptors that can bind to cell surface ligands, extracellular matrix (ECM) ligands and soluble ligands. Integrins have two main biological functions, including attaching the cell to the ECM and transducing the biological signals from the ECM to the cell. In addition, integrins are also involved in a large variety of other biological functions, such as immune patrolling, cell migration, and binding to cells by certain viruses (e.g. adenovirus, echovirus, hantavirus, and foot and mouth disease viruses).

Among Mempro™ cell-free membrane protein production systems, E. coli S30 extract is widely used for integrin production. Wheat germ extract can provide eukaryotic translation system for integrin expression. Baculovirus expression system from Autographa californica nucleopolyhedrovirus (AcNPV) is also a major system for integrin expression. Rabbit reticulocyte lysate is another useful tool for eukaryotic cell-free transmembrane protein expression. To select an appropriate source of the extract, many factors, such as the availability of materials, protein origin and complexity, downstream processing, etc., should be considered.

With the Mempro™ cell-free protein production platform, Creative Biostructure are capable of expressing, purifying and crystallizing integrins to perform further structural and functional studies.

One challenge associated with cell-free protein production is that degradation of the DNA by endogenous nucleases. Creative Biostructure can optimize Mempro™ cell-free protein production for integrins using plasmids due to they are circular and have no end for the exonucleases to attach.
