Mempro™ Cell-Based Annexins Production

Mempro™ cell-based protein production system is widely used for the production of membrane proteins. Annexins are a well-known group of membrane binding proteins, which are Ca2+-regulated phospholipid binding proteins. Members of annexin family contain four conserved structural elements termed annexin repeats composed of approximately 70 amino acid residues. Annexin repeat is highly α-helical and slightly curved disc that has a surface containing the Ca2+- and membrane-binding sites. It has been reported that annexins play important roles in vesicle transport (such as exocytosis and endocytosis) and calcium ion channel formation. Additionally, annexins can function as membrane scaffolding proteins to anchor other proteins to the cell membrane.

Escherichia coli (E. coli) is the most commonly used bacterial host for the production of annexins . Yeast cells (such as saccharomyces cerevisiae and pichia pastoris) system, combines prokaryotic as well as eukaryotic characteristics. It is a powerful eukaryotic host due to single cells, fast growth rates, inexpensive media, high cell densities. Insect cells (such as Sf9, Sf21 and High Five) are also the major system for annexins expression, which are developed from transfected insect cells in combination with vectors derived from the baculovirus species AcMNPV. Mammalian cells present another eukaryotic system for annexins expression, which provide near-native cell environment. Cell lines derived from COS, CHO, BHK-21, HEK293, HeLa and GH3 can be widely used for membrane protein expression.
