Mempro™ Detergent-Free Receptor-Type Kinase Production

Inquiry

Creative Biostructure can provide unparalleled custom Mempro™ receptor-type kinase production services using detergent-free expression system.

Receptor-type kinases are a superfamily of high-affinity and membrane-spanned receptor proteins that play a crucial role in signaling transduction. Receptor-type kinase contains a single hydrophobic transmembrane-spanning domain, however, most receptor-type kinases are single subunit receptors but some exist as multimeric complexes. Notably, as the ligands bind to the extracellular domain of receptor-type kinase, which can induce formation of receptor dimers. Conventional approaches for receptor-type kinase production are carried out through extraction from the membrane in the presence of detergent. However, detergent can induce changes on membrane protein conformation. Thus, stable solubilization and native conformation of receptor-type kinases are important issues in biotechnological field. Our alternative Mempro™ detergent-free expression system can best preseve the native conformation of receptor-type kinases for further biological studies.

Creative Biostructure have great expertise in high-yield receptor-type kinase production using detergent-free membrane protein expression system, we can perform various strategies for Mempro™ detergent-free receptor-type kinase production, including:

  • Mempro™ receptor-type kinase production using nanodiscs

Nanodiscs are the self-assembly nanotechnology to stabilize membrane proteins removed from the membrane by membrane scaffold proteins (MSPs) in no need of detergent. Creative Biostructure can perform recombination of receptor-type kinase expressed by cell-free expression system with pre-assembled nanodiscs in absence of detergent.

  • Mempro™ receptor-type kinase production using amphipols

The amphipathic amphipols have the ability to enwrap desired membrane proteins, allowing them to stay folded. Creative Biostructure can apply A8-35 to solubilize receptor-type kinase during purification process.

  • Mempro™ receptor-type kinase production using poly (styrene-co-maleic acid) lipid particles (SMALPs)

The SMALPs are self-assembled by the simple addition of the SMA co-polymers. At neutral or alkaline pH, the disc-like structure assembles itself, encapsulating the receptor-type kinase in a form amenable to be purified.

These novel detergent-free technologies for receptor-type kinase production can be obtained easily, and enabling more comprehensively structural and functional studies.

Creative Biostructure can offer other various Mempro™ membrane protein production services. Please feel free to contact us for a detailed quote.
