The major facilitator superfamily (MFS) enables to accelerate the movement of diminutive solutes get through cell membranes, so they can response to chemiosmotic gradients. However, MFS is a class of membrane transport proteins, which are expressed widely in all part of life for the import or export of target substrates. In the premier researches, scientists regarded the MFS family as proteins mainly uptake of sugars, while subsequent studies showed that oligosaccharides, amino acids, metabolites, drugs and oxyanions are all in the region MFS transported. The initial structures of the major facilitator superfamily which have been solved were glycerol 3-phosphate/phosphate exchanger GlpT and the lactoseproton symporter LacY. These contribute to clarify the overall structure of the protein family and provide initial models for understanding the MFS transport mechanism.
