Peptidase AD, also called gamma secretase complex, which plays an important role in the progress of Alzheimer’s disease (AD). Peptidase AD is an 18-spanning transmembrane aspartyl protease that can regulate intramembrane proteolysis for a growing list of integral membrane proteins. The most famous cleavage substrate of peptidase AD is amyloid precursor protein, which is closely related to AD. It has been shown that peptidase AD superfamily is composed of three protein families: Type IV leader peptidase, Presenilin and Gamma-secretase. Gamma-secretase family contains seven members, including Gamma-secretase complex, structure 1; Gamma-secretase complex, structure 2; Gamma-secretase complex, structure 3; Gamma-secretase complex, structure 4; Gamma-secretase complex, structure 5; Gamma-secretase complex, structure 6 and Nicastrin, TM helix. Up to now, several drugs act as gamma-secretase inhibitors have been developed to treat Alzheimer’s diseases.
