Mempro™ cell-free protein production systems are the innovative systems to produce a wide range of integral membrane proteins (including PH domain-like superfamily proteins), which can bypass the limitations that encountered in cell-based protein production systems, such as host cell toxicity, low yield and solubilization and purification using detergents (Figure 1). PH domains are the small molecular domains that can bind phosphoinositides with high affinity and specificity. The significant property of PH domain is that this specific domain can introduce phosphorylation at different sites within the inositol ringbind lipid. In other words, PH domains have lipid binding specificity.

Baculovirus expression system from Autographa californica nucleopolyhedrovirus (AcNPV) is also a major system for PH domain-like protein expression, which is developed in the early stage that cultivated insect cells in combination with the baculovirus vector (bacmid). Crude rabbit reticulocyte lysate is also the most widely used for eukaryotic cell-free membrane protein expression systems.
