GLTP is a 24 kDa protein presents in many species that catalyzes the transfer of glycolipids through the intracellular membranes. Both sphingoid- and glycerol-based glycolipids can be transferred by GLTP as long as the hydrophobic lipid backbone has beta-linked initial sugar. The activity of GLPT depends on the composition and the properties of the membranes the glycolipid exists, such as the packing of the membrane and whether the membrane consists of sphingomyelin or phosphatidylcholine. All the known GLPTs from different organisms showed a 3-D structure of a double-layered all-α-helical conformation with a ligand-binding site. The binding site consists of a sugar recognition site and a channel for hydrocarbon chain binding. The interaction between glycolipid and GLTP arises from the hydrogen binding and the hydrophobic effect at the binding site. This unique structure differentiates GLTP from other known lipid transfer proteins.
