Mempro™ Cell-Free Saposin Production

Inquiry

Creative Biostructure offers advanced custom Mempro™ protein production services for saposine using cell-free expression system.

Mempro™ cell-free protein production system is a robust and convenient system developed by Creative Biostructure for efficient production of membrane proteins. It holds many advantages over the traditional cell-based protein production system, including less concern of toxicity, high yield, ready-to-use post-translational modifications etc.

Saposins are small glycoproteins that activate lysosomal hydrolases that participate in metabolism of sphingolipids. Mature saposins are derived from prosaposin, a common precursor molecule. The structures of all the four types of mature saposins (A, B, C, D) are similar, containing six equally placed cysteines, two conserved prolines at the same locations and a glycosylation site. Saposins have highly compact structures which results in its high heat-stability and strong resistance of the protease attack. α-helices are conserved in saposin structure, and are maximal at pH 4.5 which is the optimal pH for lysosomal hydrolases. In addition, the first twenty-four amino acids may assign a β-sheet configuration at N-terminus of saposins.

Creative Biostructure is very experienced in high-yield Saposin production using cell-free protein expression systems, various strategies are available for Mempro™ cell-free protein production, including:

  • Mempro™ Cell-Free Protein Production in E. coli;
  • Mempro™ Cell-Free Protein Production in Wheat Germ;
  • Mempro™ Cell-Free Protein Production in Rabbit Reticulocyte;
  • Mempro™ Cell-Free Protein Production in Baculovirus.

Among Mempro™ Cell-Free Protein Production systems, baculovirus system is the best for saposin production. Baculovirus expression system was derived from Autographa californica nucleopolyhedrovirus (AcNPV). It’s particularly suitable for large scale application. It’s proved that the baculovirus system produces abundant and active saposin protein.

Creative Biostructure is a leading expert in cell free protein expression. With the Mempro™ cell-free protein production platform, purified saposin could be produced in a cost-effective manner, which will ensure the progress of your research.

We also provide other types of Mempro™ membrane protein production services. Please feel free to contact us for a detailed quote.
