Creative Biostructure can provide unparalleled custom Mempro™ immunoglobulin-like superfamily (E set) protein production services using detergent-free expression system.

Immunoglobulin superfamily (IgSF) is a large group of membrane-bound proteins that are involved in the processes of recognition, binding, and adhesion to cells. Members of IgSF contain a sturctural domain termed Ig domain, which possess around 70-110 amino-acid residues. Immunoglobulin-like superfamily is also termed E set, including a wide range of domains that have an Ig-like fold in anti-parallel β-strands. Conventional approaches for E set protein production are performed through extraction from the membrane in the presence of detergent. However, detergent can induce conformational changes on membrane protein. Thus, stable solubilization of E set proteins is important issue in biotechnological field. Our alternative Mempro™ detergent-free production system can best preseve the native conformation of E set proteins for further biological studies.

  • Mempro™ E set protein production using nanodiscs

Nanodiscs are the self-assembly nanotechnology to stabilize membrane proteins removed from the membrane by membrane scaffold proteins (MSPs) in no need of detergent. Creative Biostructure can perform combination of cell-free expressed E set proteins with pre-assembled nanodiscs in absence of detergent.

  • Mempro™ E set protein production using amphipols

The amphipathic amphipols have the ability to enwrap desired membrane proteins, allowing them to stay folded. Creative Biostructure can apply A8-35 to solubilize E set proteins during purification process.

  • Mempro™ E set protein production using poly (styrene-co-maleic acid) lipid particles (SMALPs)

The SMALPs are self-assembled by the simple addition of the SMA co-polymers. At neutral or alkaline pH, the disc-like structure assembles itself, encapsulating the E set proteins in a form amenable to be purified.

These novel detergent-free technologies for E set protein production can be obtained easily, and enabling more comprehensively structural and functional analyses.
