The major facilitator superfamily (MFS) is a family of membrane transport proteins that transport small substrate, such as sugars, amino acids and metabolites,in response to chemiosmotic gradients. They are found to be important in three kingdoms. MFS is a single polypeptide protein that consists of 12 transmembrane helices at N-terminus. two 6-helix bundles at C-terminus and connected by the cytoplasmic loop. The transmembrane helices and the extracellular loops have strong interactions with each others at the ends, which seal the two halves of the protein against each other, forming a clam-shell. The unique structure allows an open space at the membrane, to cytoplasm or periplasm/extracellular space. The specificity of transportation was determined by the amino acids at the cavity. In addition, many studies showed that MFS may form a dimer for its function in the cell.
