What is cyclodextrins (CDs)?

Cyclodextrins (CDs) are chemically and physically stable cyclic oligosaccharides produced by enzymatic degradation of starch. Due to the chair conformation of glucopyranose, CDs maintain the shape of truncated cone or torus. (Figure 1). CDs are characterized by hydrophilic outer surface and lipophilic cavity, thus they form water-soluble inclusion complex with a wide variety of hydrophobic compounds and change physicochemical and biological properties of guest molecules. Therefore, cyclodextrins has huge potential to be exploited in a wide range of applications in food, pharmaceutical and chemical industries, agriculture and environmental engineering.

Cyclodextrins are classified as natural and cyclodextrins derivatives. Cavity size varies depends on how many glucopyranose units compose the rings. Natural cyclodextrins are 𝛼, 𝛽, and 𝛾 consisting of 6, 7, and 8 glucopyranose units have been commonly used, as well as their derivatives. The ability of a CD to form an inclusion complex is related to factors involving cavity size, solubility, binding affinity, and availability, etc. Various hydrophilic, hydrophobic, and ionic derivatives have been developed and utilized to modify its physicochemical and biopharmaceutical properties, and then meet the drug delivery need with improved inclusion capacity of natural cyclodextrins.
