Why cyclodextrins are used for drug delivery?

The molecules associated with poor physicochemical and biopharmaceutical properties are difficult to formulate.   • Cyclodextrins in drug delivery systems: Cyclodextrins could be used as drug carriers like liposomes, it has advantages in different drug delivery route including ophthalmic, nasal transdermal, rectal and oral drug development. Cyclodextrin modification could achieve increased solubility for a specific drug molecule. More importantly, double loaded technology (eg. Drug-in-Cyclodextrin-in-Liposome) combines relative advantages of CDs and liposomes into one system to prolong drug release process, increase target release, and significantly enhance drug-loading capacity. In particular, this double loading system is suitable for the delivery of hydrophobic drugs which also possess volatility. Besides liposome, this method also could be combined with other nanocarriers.
  • Cyclodextrins enabled injections: Cyclodextrins used in injectable formulations have advantages with desirable physical and chemical stability, isosmotic property, pH close to interstitial liquid over formulations that contain detergents like TWEEN or co-solvents.
  • Cyclodextrins in biological products: Cyclodextrins are used as replacements for traditional and less safe excipients (e.g. detergents) in formulations of highly concentrated antibodies to overcome associated challenges in terms of optimization of production processes, chemical and physical stability of the final products, preventing aggregation and solution viscosity.
